SUMMARY
Cochrane systematic review assessing the alleged anti-cancer effect and possible adverse effects of laetrile and amygdalin, searching eight databases and two trial registers. The review's conclusion, per its indexed abstract: the claims that laetrile/amygdalin benefit cancer patients are not supported by sound clinical data, there is a considerable risk of serious adverse effects from cyanide poisoning (especially after oral ingestion), and the overall risk-benefit balance is unambiguously negative. PMID 22071824 confirmed via PubMed/NCBI eutils esummary and Crossref DOI resolution; abstract wording per EXA-indexed abstract text, not independently re-fetched from a directly loaded PubMed page (PubMed's page presented a cookie-consent interstitial to automated fetch during this research session).
NOTES

Current, methodologically rigorous secondary confirmation (Cochrane) that the disconfirmation is not a one-off 1982 finding but the settled position of a systematic review of all available clinical evidence.
