MECHANISM CLAIMED
Uncouples oxidative phosphorylation in mitochondria: protons that would normally drive ATP synthase leak back across the inner mitochondrial membrane, so cellular energy is dissipated as heat instead of captured as ATP. This genuinely raises metabolic rate and burns fat, but the same mechanism produces uncontrolled heat generation once a small threshold is crossed, and there is no antidote; the therapeutic and lethal doses lie close together.

2,4-Dinitrophenol is a yellow, industrially produced nitrophenol whose capacity to uncouple mitochondrial oxidative phosphorylation was first exploited as a weight-loss treatment in the United States beginning in 1933. The mechanism is real and remains scientifically uncontroversial: DNP does increase metabolic rate and does burn fat. What makes it uniquely dangerous is the absence of any meaningful safety margin between an active dose and a fatal one, and the absence of any antidote once toxicity begins. The U.S. Food and Drug Administration declared it “extremely dangerous and not fit for human consumption” in 1938 and discontinued its medicinal use (ATSDR); in the United Kingdom it is not sold as a licensed food or medicine, and regulators have found DNP in diet products and prohibited DNP-containing products (FSA, 2003; Dawson et al., 2025). It resurged as an unlicensed grey-market chemical, imported industrially and repackaged into capsules for dieters and bodybuilders: a 2019 U.S. federal toxicology profile recorded it was still marketed by unregulated internet sources, with DNP deaths reported in the medical literature as recently as 2024.
